Abstract

This study aimed to investigate the modulating effects of rutin against hepatotoxicity of MSG on rats through measurement of lipidperoxidation, total antioxidant, Bcl-2, caspase-1, DNA fragmentation and comet assay .60 male rats divided into 6 groups which are , the first group (control group) , the 2nd and 3rd administrated with 25 , 50 mg/kg b.wt of Rutin, respectively , the 4th group administrated with 8 mg/kg b.wt of MSG .The 5th and 6thgroups administrated with MSG + Rutin ( 25 ,50 mg/kg b.wt ), respectively , all administrations were orally gavage for 4 successive weeks. A significant increase in lipid peroxidation was showed, while there is a reduction in total antioxidant activity in MSG group, However , the two doses of Rutin improved these disorders. MSG caused down regulation of Bcl-2 protein and increases the caspase-1 level ,while treatment with Rutin increased the Bcl-2 content and reduced caspase-1 level. MSG only showed faint , smear genomic DNA and an increase in DNA strand breaks in the hepatic tissue cells. However, treatment with Rutin 25 mg or 50 mg enhanced these disorders .In conclusion, our results confirmed the effective role of rutin against MSG induced hepatotoxicity in male rats.
